INCUBATION OF THE EXPLANT

The cultures are incubated in an air conditioned room at a constant temperature of 25 C.for temperate plants and 27 C for tropical plants. They are kept on lighted shelves with fluorescent lights spaced 20 inches apart. To prevent excessive heat build up remove the chokes from the tube lights and wire them outside the room. Instead of tube lights you can use CFL lamps also. The lights should be on for 16 hours and off for 8 hours

The incubation room or growth room should be kept thoroughly clean. Wash the floor every day with a disinfectant solution.
